Modifications of hemolytic activity of polyene antibiotics by mono- and disaccharides in mammalian erythrocytes

The kinetics of the filipinand amphotericin-B-induced hemolysis of pig, horse and human erythrocytes in isotonic KCl, monoand disaccharide media was investigated. The kinetics of filipininduced hemolysis is of the damage type, with small changes in erythrocyte resistance to the antibiotic as the standard KCl medium is replaced by the saccharide media. The hemolysis induced by amphotericin B is highly differentiated. The kinetics of the hemolysis in KCl medium is of the permeability type. In monosaccharide media it is changed to the damage type, with a small increase in erythrocyte resistance to the antibiotic (about 2.5 times). In disaccharide media, the resistance to amphotericin B is highly increased (about 2 orders of magnitude), but the kinetics of hemolysis is of the permeability type, as in KCl medium. In conclusion, amphotericin B molecules form in the membranes several types of transporting complexes with properties depending on antibiotic concentration, chemical composition of incubation medium, and small differences in membrane organization.
